Tumor necrosis factor-alpha release by human peripheral blood mononuclear cells treated in vitro with anticancer drug etoposide

Abstract

Mononuclear cells (MNCs) from human peripheral blood were cultured for 20 hours with or without etoposide (5.0, 50.0 or 250.0 pg per ml). Dying MNCs (in a stage of secondary necrosis) as propidium iodide (PI) - positive cells and a level of tumor necrosis factor - alpha (TNF-a) in cell supernatant were counted after termination of culture period. It was shown that high (250.0 pg per ml), but not low doses of etoposide induced substantial increase the number of Pl-positive cells and the level of TNF-a in the supernatant of cultured MNCs of healthy people and oncological patients. We concluded that TNF-a release is an indication of cell necrotic death, induced by anticancer chemotherapeutic agent etoposide.
