MORPHOLOGICAL CHARACTERISTICS AND THE COMPOSITION OF TYMUS CELLS IN EXPERIMENTAL ACUTE ULCERATIVE COLITIS OF DIFFERENT SEVERITIES

Abstract

The morphological changes of thymus and its cell composition were studied in experimental acute colitis of variable severity - catarrhal-ulcerative and fibrinose-ulcerative, induced with 1% and 5% water solution of sodium dextran sulfate salt. Annexin V-positive cells, which die by apoptosis, were identified in the thymus using flow cytofluorimetry. The chromogenic LAL-test was used to evaluate the endotoxin level in the blood serum. Compared to the control group the serum endotoxin level was 3.9 time higher in the mice with catarrhal-ulcerative colitis, and 9.6 times higher in the mice with severe fibrinose-ulcerative colitis. During acute catarrhal-ulcerative colitis in the adult male C57Bl/6 the accidental thymic involution is moderate, while in the mice with fibrinose-ulcerative colitis it is pronounced. During the moderate and severe accidental thymic involution the number of dying cells in the thymus increases 1.2 and 1.8 fold respectively, there is a decrease in the lymphocyte total number, CD3-CD19+ В-, CD3+CD8+ cytotoxic, Т-, CD4+CD8+ double positive lymphocytes, CD3+CD4+ Т helpers and CD4+CD25+Foxp3+ regulatory T lymphocytes, F4/80+ macrophages. During the moderate accidental involution the content of CD4-CD8-CD45+CD11c+dendritic cells decreases in the thymus and in the severe accidental involution there is an increase of the content of CD4-CD8-CD45+CD11c+dendritic and CD326+UEA+CD205-epithelial cells in the thymic medulla.
