ANALYSIS OF FOXP3 ISOFORMS EXPRESSION BY REGULATORY T CELLS FROM PERIPHERAL BLOOD IN ALLERGIC RHINITIS BEFORE AND AFTER SPECIFIC IMMUNOTHERAPY

Abstract

We have analyzed the number of regulatory T cells and the level of FOXP3 isoforms expression by flow cytometry and mRNA expression of the transcription factors GATA3, TBX21 and RORC by real-time PCR in peripheral blood of patients with seasonal allergic rhinitis (or rhinoconjunctivitis). Before the therapy, there was e a Th2/Th1 imbalance (increase in the ratio of GATA3/TBX21) and a decrease in the total number of regulatory T cells mainly due to amount reduction of regulatory T cells expressing FOXP3 molecule lacking exon 2. The last fact we have estimated as a possible decrease in the functional activity of regulatory T cells. After specific immunotherapy we observed a relative increase in the functional activity of regulatory T cells through the accumulation of cells expressing FOXP3 molecule lacking exon 2, a decrease in the GATA3/TBX21 ratio and a lower level of RORC mRNA expression. Those changes had long-term character, remaining at least until the end of the pollination season, and we have assessed them as a positive effect of the specific immunotherapy.
