COMPARATIVE ANALYSIS OF PROINFLAMMATORY AND TOLEROGENIC FACTORS EXPRESSION IN HUMAN MESENTERIC HEMAL AND LYMPH NODES

Abstract

The mesenteric group of human lymph nodes is known for its active participation in the induction of immunological tolerance and contains single hemal nodes. A distinctive feature of these nodes is blood circulation in the sinus system, although the functional value of this feature remains unclear. In our study, we compared the expression of genes of tolerant factors FOXP3, TGF-β, IL-10 and transcription factors of proinflammatory («colitogenic») forms of T-helpers - RORC (Th17-cells), TBX21 (Th1-cells) in human mesenteric hemal and lymph nodes using real-time PCR. As a result, it was found that the level of the relative normalized number of mRNA genes of tolerant factors is reduced in the hemal nodes compared to lymph nodes, and the transcription factors of proinflammatory forms of T-helpers are increased. At the same time, a greater number of neutrophilic granulocytes were detected in the sinus system of the hemal nodes compared to the peripheral blood, which may be an indirect confirmation of the presence of Th17 cells. Summing up the data obtained, it can be concluded that compared with mesenteric lymph nodes, the hemal nodes are characterized by a shift in the balance of tolerant/proinflammatory factors towards the latter, which makes it promising to conduct further studies of mesenteric GU in inflammatory bowel and liver diseases.
